This kit uses EFC technology to measure ligand-induced internalization of CHRM2 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged CHRM2. The detection and measurement of free thiols (i.e., free cysteine, glutathione, and cysteine residues on proteins) is one of the essential tasks for investigating biological processes and events in many biological systems. Cayman’s Thiol Detection provides a simple, reproducible, and sensitive tool for assaying free thiol content in samples (i.e., plasma, serum, tissue homogenates, cell lysates, and urine). The assay utilizes a proprietary fluorometric detector that reacts with thiol groups to emit a strong fluorescent signal which can be detected using excitation wavelengths between 380-390 nm and emission wavelengths between 510-520 nm.
Montana Molecular Borealis Arrestin AT1R Assay - 10mL
Measure arrestin signaling kinetics in real time with the live cell, fluorescent Borealis arrestin - Angiotensin II Type I receptor (AT1R) assay. Add the unmodified AT1R and Borealis sensor to cells and measure robust signals the next day on an automated plate reader or fluorescence imaging system. This assay is genetically encoded and packaged in BacMam for efficient transduction in most cell types - including uniform and near 100% efficiency in HEK293 cells as well as expression in many primary or iPSC-derived cells. The kit includes the arrestin sensor BacMam, AT1R BacMam, 500mM sodium butyrate in H2O, and a positive control receptor and agonist (AT1R BacMam and 5mM Angiotensin II in H2O). Borealis kits for several other receptors are also available, and larger-sized kits are available on request. Other Montana Molecular tools including the DAG and Calcium sensors can be used alongside this assay.
